低海拔香菇品系之育種

Studies on the Breeding of Shiitake-A New Strain for the Cultivation in the Low Elevation Areas

摘要


由臺灣採集之野生香菇5種,取其子實體所得之單核菌絲各10個,與4種日本野生品系及6種日本栽培品系進行什交試驗,共得5×10×10個什交組合。經培養後檢查其扣子體產生情形,得到162個什交雙核體可產生扣子體,再進行菌絲培養試驗及低溫下產生原基體能力之測定,最後得到24個組合。以此爲原種,製作成菌種,並於低海拔地區進行試種,大部份組合體均不能適應高溫下之生長,僅有K53×D54(L107)可正常生長,且產量高,其特陸爲本菌株可以在高溫之低海拔下栽培,子實體形狀大,產量多,並可作爲局部地區推廣及進一步育種之材料。本試驗證實什交育種可作爲一種解決本省香轄品系來源之途徑,以得到適合本省栽培之高產優良品系。

In this study, five strains of Lentinus edodes from Taiwan, 4 wild strains and 6 cultivated strains from Japan were used in the di-mon mating method to breed a new strain for the cultivation in the low elevation areas in Taiwan. Ten single spore isolated each of the five wild strains from Taiwan were used as monokaryons and both wild and cultivated strains from Japan were used as dikaryons which resulted in 500 all possible crosses, isolates or di-mon matings. Twenty-four out of the 500 isolates produced primo rdia on agar medium with a strong vigor of mycelial growth. The cultivation trails of the 24 isolates at Kuan-tien (25m from sea level) indicated that the isolate L107 (K53×D54) was very promising because it produced an economic yield and the shape and size of its fruiting bodies were normal and large. From the large scale trials on different kinds of logs at different locations in the areas of less than 250m from sea level, isolate L107 produced economic yield and the quality of the fruiting bodies is also good. Therefore, it can be either cultivated in the low elevation areas in Taiwan, or used as a breeding material for further improvement.
