利用RAPD分子標誌估測番椒屬種原遺傳歧異性之研究

Genetic Diversity within Capsicum Evaluated by Random Amplified Polymorphic DNA (RAPD) Markers

摘要


本試驗以蒐集之番椒屬(Capsicum)44個種原,含5個馴化栽培種(C. annuum, C. baccatun, C. chinense, C. frutescens, C. pubescens)與3個野生種(C. chacoense, C. praetermissum, C. eximiun),探討利用逢機增殖多型性DNA(RAPD)標誌,進行「種」的鑑別與遺傳歧異性之研究。試驗結果,以UBC327, UBC346, UBC457, UBC483, UBC484,等五個逢機引子,經聚合酵素連鎖反應(PCR),其RAPD產物具有明顯的多型性表現,可用以區別5個馴化栽培種。將上述所有多型性的DNA片段一併進行Neighbour Joining method群集分析,結果44個品系可分成六群:第一群有7品系,主要為C. chinense;第二群有7品系,含C. praetermissum、C. eximium與C. pubescens;第三群有3品系,主要為C. chacoense;第四群包含所有的C. baccatum;第五群主要包含所有的C. frutescens;第六群則主要包含全部之C. annuum。試驗結果顯示利用RAPD標誌之分析,可與「種」為基礎之分類吻合。

A total of 44 collections of pepper (Capsicum.spp.) germplasm, including five domesticated species (C. annuum, C. baccatum, C. chinense, C. frutescens, C. pubescens), and three wild species (C. chacoense, C. praetermissum, C. eximum), were evaluated using random amplified polymorphic DNA (RAPD) marker. Objectives were to assess genetic diversity within the germplasm of Capsicum and survey the identification markers. The results indicated that high degree of polymorphism was obtained from five random decamer primers (UBC327, UBC346, UBC457, UBC483, and UBC484) which produced 172 RAPD bands, and within them ten efficient fragments useful for identification among five domesticated species had been screened. Analysis of similarity by NJ (Neighbour Joining method ) revealed these 44 accessions could be divided into six groups: the first group contained seven accessions that including all the samples of C. chinense; the second group was composed of seven accessions, including the species of C. praetermissum, C. pubescen and C. eximum; the third group contained three accessions belonging to C. chacoense; , the fourth group contained all the samples of C. baccatum the fifth group contained all the samples of C. frutescens; , and the sixth group contained all the samples of C. annuum. The results showed that the clusters assorted by RAPD markers could be corresponded to species-based classification.
