摘要


溶血現象為病鰻愛德華氏菌(Edwardsiella tarda)或嗜水性產氣單胞菌(Aeromonas hydrophila)感染所顯現的重要病徵之一。愛德華氏菌的溶血基因已由ET16菌株分離出並轉殖入pSK載體,於大腸桿菌轉形宿主細胞內表現溶血毒素。所選殖的溶血基因核苷酸片段估計為5.3 kb長度。以已知的嗜水性產氧單胞菌溶血基因(aerolysin)做探針檢測,核苷酸雜交的結果顯示兩種水生病原菌的溶血基因完全不同。所選擇的溶血基因於大腸桿菌轉形宿主細胞內所表現的溶血毒素估計為34 kDa,與以愛德華氏菌ET16菌株的細胞外培養液,於聚丙醯胺膠作溶血毒素活性檢測,估算溶血毒素的大小為34 kDa一致。又經由次選殖轉形篩選,其溶血活性喪失,可確定EcoRI位於溶血基因內。因此該攜有愛德華氏菌溶血基因的轉殖重組質體命名為pETH3E。
