Efficient Identification of Vibrio cholerae by PCR Using sodA Sequence and Serogroup RAPD Differentiation

以霍亂弧菌(Vibrio cholerae)超氧歧化酶基因(Vc sodA)PCR結合隨機複製多型性DNA (RAPD)技術有效鑑別霍亂弧菌及其血清型

摘要


本研究已研發出快速有效鑑別霍亂弧菌(Vibrio cholerae)及其血清型之方法。利用霍亂弧菌超氧歧化酶基因(superoxide dismutase gene, Vc sodA)作為該菌種之鑑別,再結合隨機複製多型性DNA (Random Amplified Polymorphic DNA, RAPD)技術,結果顯示弧菌染色體DNA以Vc sodA基因序列進行PCR,可生成霍亂弧菌特異性之產物615 bp DNA,RAPD分析可以明確區別霍亂弧菌不同的血清型。本研究提供了一個可靠、有效而迅速的霍亂弧菌血清型鑑別方法。

The Vibrio cholerae superoxide dismutase gene (Vc sodA) was used as the target for species identification. Results revealed that Vc sodA sequence-based PCR specifically amplified a 615 bp DNA fragment from V. cholerae chromosomal DNA, whereas no PCR DNAs were produced from other Vibrio species tested in this study. The rapid amplification of polymorphic DNA analysis could clearly distinguish serotypes such as O1, Inaba (+/-), Ogawa (+/-), O139, and NonO1 of V. cholerae under the conditions presented in this study. This combination method appears to be valuable for the rapid identification of V. cholerae and the differentiation of its serotypes among Vibrio species.
