戴奧辛細胞檢測系統之建立

Establishment of Cellular Detection Systems for Dioxin-like Compounds

摘要


戴奧辛(dioxin)是一種苯環類化合物之統稱,也是一種著名的環境毒物,由於具有高度脂溶性,故常經由食物鏈途徑在生物體內積存,對其生理系統會造成很大的影響,甚至是細胞組織的病變。目前國內外針對戴奧辛的檢測方法仍是以化學分析方法為主流,費時且耗費昂貴,隨著人們對戴奧辛在生物體內致毒機制了解的日益增,以生物反應為基礎的生物檢測方法(bioassays)逐漸受到重視。目前戴奧辛之生物檢測方法包括了in vivo biomarkers, in vivo bioassays,和in vitro bioassays三大類。然而In vitro bioassays系統較能真實反應戴奧辛對於生物體的毒害程度,且在時間及金錢上佔有相對的優勢。有鑒於戴奧辛生物檢測對戴奧辛生物危害的直接意義,本研究期望可以利用細胞反應來辨識戴奧辛環境污染物,因此我們以CALUX (chemical-activated luciferase expression) assay為基礎,建立含有dioxin response element和luciferase gene的表達質體,分別送入不同的細胞株中,結果發現冷光的表現和戴奧辛含量有明顯正向關係,希望藉由此冷光的定量做出一組標準曲線圖,使其更進一步成為比現今檢測方法更為省時有效的環境污染物檢測工具。

Dioxins are group of compounds contain double aromatic ring-like structure. They are among the most prevalent and toxic environmental pollutants. Accumulation of dioxins in human tissue poses a potential threat to human health. Currently, analytical chemical procedures dominate the dioxin detection protocols. However, these procedures are both expensive and time-consuming, which preclude their used in routine monitoring. Assays employing biological methods, including in vivo biomarkers, in vivo bioassays and in vitro bioassays, offer a powerful alternative to chemical procedures. These methods based on biological effects are detecting 'toxicological specificity', which refers to the relationship between the assay response and the toxic potentiality rather than simply the concentration of contaminants in the sample being analyzed. This report reviewed the current biological detction methods for dioxins and also reported the establishment of a cell-based reporting system to identify dioxin-like compounds.
