由紅藻絲狀體產製藻紅素研究

A study on the production of phycoerythrin from the conchocelis of Rhodophyceae

摘要


本計畫利用紅藻絲狀體為原料,開發產製天然紅色色素的來源,試驗所使用的絲狀體材料分別為:蠕枝藻(Helminthocladia australis Harvey)、狹葉紫菜(Porphyra.angusta Ueda)、頭髮菜(Bangia atropurpurea(Roth)C. Agardh),各項試驗所獲得之成果如下:一、三種不同紅藻絲狀體中以蠕枝藻(Helminthocladia australis Harvey)的生長速率及藻紅素含量較有開發潛力,目前的生長速率為1.9g/l/m,藻紅素含量為18.3%。二、紅藻絲狀體生長速率改善:利用田口式品質工程法找出較佳之藻種為蠕枝藻(Helminthocladia australis Harvey),初步定出適宜之生長條件為:添加維他命B_(12) 1ug/l,NaNO_3 0.22 g/l,Na_2HPO_4 22 mg/l,溫度28°C,鹽度3.5%,光強度2000 lux,光照週期12hr/day。三、藻紅素萃取分離純化:以利用珠磨機研磨最為有效,抽取率可高達80%以上。利用加熱方式可去除藻萃取液中之藻藍素,但效果只達到50%。若用膜分離方法,則可使藻紅素的純度提高2.4倍,但仍有61%的損失。四、藻色素實驗工廠規劃設計:根據養殖、萃取、濃縮、乾燥、純化等各項試驗資料,規劃藻色素實驗工廠之簡易加工試產線。

The present study investigated the conchocelis of Rhodophyceae as a source of natural pigments. There were three conchocelis of Rhodophyceae, including Helminthocladia australis Harvey, Porphyra.angusta Ueda, and Bangia atropurpurea(Roth) C. Agardh, being used. The conchocelis from Helminthocladia australis Harvey was found to give best results, with a growth rate and phycoerythrin content of 1.9g dry matter/l/month and 18.3% respectively. The optimal growth condition for the conchocelis of Helminthocladia australis Harvey, established by Taguchi method, was: Vitamin B_(12) 1 ug/l, NaNO_3 0.22 g/l, Na_2HPO_4, 22 mg/l, temperature 28 °C, salinity 3.5%, light intensity 2000 lux, and a light period of 12hr/day. It was also found that more than 80% of the phycoerythrin could be extracted using a Dyno mill and heating could only remove about 50% of the phycocyanin within the algae extract. The purity of the extracted phycoerythrin could be improved by 2.4 times using membrane diafiltration process, this was however accompanied by a 61% lost in yield. According to the present results on culture, extraction, separation, concentration, and purification, the basic design of a pilot plant process for obtaining phycoerythrin was drafted.
