植物生長調節劑對甘藷組織培養生産花青素之影響

Effects of Plant Growth Regulators on Anthocyanin Production in Tissue Culture of Ipomoea batatas L.

摘要


本研究採用甘藷之莖、葉及塊根爲試驗材料,利用組織培養法探討生産天然色素之可行性。結果顯示,培養兩週後葉片可産生紫紅色的癒傷組織,而莖及塊根則無此現象。auxins及cytokinins對葉片培植體誘發癒傷組織之效果,以不同組合之2,4-D+kinetin或NAA+BAP進行試驗。結果顯示添加2,4-D及kinetin會促進紫紅色癒傷組織産生,NAA及BAP則無此效果。其中以1μ M 2,4-D+1μ M kintin或1μ M 2,4-d+0.1μ M kinetin效果較佳。

Stems, leaves and tuber roots of Ipomoea batatas L. were used as experimental materials in this study to investigate the possibility of production of anthocyanins by tissue culture methods. Resnlts showed that leaf explants yielded purple calli white stem and tuber root explants did not after two weeks of culturing. In order to study the effects of auxins and cytokinins on initiation of pruple calli from leaf explants of Ipomoea batatas L., different combinations of auxins and cytokinins were tested. Adding of 2,4-D and kinetin in the MS medium promoted initiation of purple calli from the leaf explants. Nevertheless, purple multi were not found on leaf explants cultured in the same medium containing NAA and BAP. Supplements of μ M 2,4-D and 1 μ M kinetic or 1 μ M 2,4-D and 0.1 μ M kinetic in the MS medium induced more purple math than the other combinations.
