光線對北蟲草生長和代謝物生成的影響

Effects of Light on Growth and Metabolite Production of Cordyceps militaris

摘要


人工栽培的北蟲草(Cordyceps militaris)需要經過長時間且週期性的光線刺激下,才會開始由白色菌絲轉為黃色菌絲,進而形成橘色長棒狀的子實體。故本研究之目的以不同的光線(紅光、藍光、白光和黑暗)去刺激北草蟲,並觀察培養皿培養、液態培養及可形成子實體之固態米基質培養下的生長和代謝物含量。結果顯示,北蟲草在固態培養皿培養下,以黑暗環境會產出最高的菌絲乾重和蟲草素含量。北蟲草液態搖瓶培養下,腺苷為一級代謝產物,在所有液態發酵初期三天的含量最高,但蟲草素是二級代謝產物,在液態發酵14天含量最高。在北草蟲固態米基質培養,以培養21天黑暗組的蟲草素最高,但培養21天各組均測不到腺苷含量。採用紅光則是助於北蟲草菌絲體的生長,然而藍光是誘導形成北蟲草子實體的關鍵光源,隨著子實體的生長,腺苷則以原基狀態含量較高,而蟲草素會隨著子實體生成而逐漸增加。

The artificial cultivation of Cordyceps militaris needs light induction with long period of time and daily cycle to turn the color of mycelium from white to yellow, then in turn forms an orange color long rod-like fruiting body. Therefor the objective of this study was used different light conditions (red, blue, white, and dark) to stimulate Cordyceps militaris qrowtth and to analyze metabolite contents in plate culture, liquid and solid-state rice cultivuation. The results showed that Cordyceps militaris produced the highest dry weight of mycelium and cordycepin in a dark environment under solid culture. Under the liquid shake flask culture of Cordyceps militaris, adenosine was the first metabolite, which was the highest contents in the first three days of all liquid fermentation. Cordycepin was the secondary metabolite which was the highest in the 14 days liquid fermentation. The 21 days dark group in the solid-state rice medium Cordyceps militaris cultivuation had the highest cordycepin content, but there was no adenosine content in each group. The red light was used to help the growth of Cordyceps militaris mycelium. However, blue light was a key source inducing the formation of Cordyceps militaris fruit body. With the growth of fruiting bodies, adenosine had a higher content in the primordium state, and cordycepin content gradually increased with the fruit body growth.
