臺灣肖楠(Calocedrus macrolepis var. formosana)是臺灣針葉五木之一,不但材質優美,且耐久性極佳。本研究目的係利用水蒸餾法萃取臺灣肖楠心材、樹皮及葉子等部位精油,計算精油收率,並評估其對紙張防黴效果之影響。各部位的精油收率,以心材的含量最高(0.54%, w/w)。比較3種精油對紙張防黴性的改善效果,以心材的防黴效果最好,對Chaetomium globosum和Aspergillus niger 2株黴菌之最低抑制濃度(MIC)分別為75µg/平方公分和400µg/平方公分,而樹皮及葉部精油則不具防黴效果。當使用濃度為100µg/平方分分時,心材精油的6個分離部(L1~L6)對C. globosum的生長皆有很好的抑制效果,當使用濃度降為50µg/平方公分時,L2、L4和L6仍可完全抑制其生長。進一步由L2分離出主要成分香旱芹酚(Carvacrol),並對2株黴菌進行紙張防黴性試驗,結果顯示,當使用濃度為200µg/平方分尺時,對A. niger的生長抑制面積為97.8%;當使用濃度為100µg/平方分分,對C. globosum的生長抑制面積為100%。綜合上述試驗結果,顯示臺灣肖楠心材精油及Carvacrol對於紙張防黴性具有良好的改善效果,非常值得進一步研究與開發利用。
Calocedrus macrolepis var. formosana is one of five precious coniferous tree in Taiwan. It has not only high quality of wood but also excellent durability. The research purposes are to extract the essential oils from C. macrolepis var. formosana heartwood, bark and leaves using water distillation and to evaluate the anti-midew property of paper treated with essential oils. Among the different parts of C. macrolepis var. formosana, the highest oil yield was obtained from heartwood (0.54%, w/w). Comparison of the anti-mildew property of treated paper, heartwood oil exhibited the best anti-mildew activity against Chaetomium globosum and Aspergillus niger with MIC values of 75 µg/cm^2 and 400 µg/cm^2, respectively. However, essential oils from bark and leaf didn't show the anti-mildew activity. Six fractions (L1~L6) of heartwood essential oil showed a great inhibitory effect on the growth of C. globosum at the concentration of 100 µg/cm^2. At the concentration of 50 µg/cm^2, L2, L4 and L6 completely inhibited the growth of C. globosum. Carvacrol was isolated and identified from L2 fraction. At the concentration of 200 µg/cm^2, it has a 97.8% inhibitory area on the growth of A. niger. At the concentration of 100 µg/cm^2, it has a 100% inhibitory area on the growth of C. globosum. Results obtained revealed that C. macrolepis var. formosana heartwood oil and carvacrol exhibited a great inhibitory effect against mildew. It is worth of further investigations and utilization.
