摘要


本試驗以青脆枝無菌種子苗之頂芽與含節莖段作為材料,建立微體繁殖技術並分析其喜樹鹼含量。結果以頂芽比節段適合作為微體繁殖材料,在培養第10週時,頂芽與節段多芽體誘導最佳培養基分別為MS添加0.25-0.5 mg/L TDZ與0.5mg/L TDZ,每個培殖體平均可產生超過20個多芽體。多芽體在MS添加低濃度BA (0.05-0.1mg/L)培養基抽長後,將單一莖芽切下培養於MS添加NAA或IBA發根培養基,以低濃度IBA或NAA (0.5 mg/L)處理最佳,可使83.3%之莖芽發根,且平均產生2.8~5.9根。來自綠島、蘭嶼與台東等不同種源芽體之多芽體增殖能力沒有差異,但發根能力則呈現明顯差異,介於22.2~100%間。調查母樹與種子苗的喜樹鹼含量,結果種源間差異很大,雖然種子苗葉片喜樹鹼含量低於母樹,但與母樹呈現正相關,根的喜樹鹼含量高於葉片,有些苗木根的濃度比葉片高出30倍。

Micropropagation and comptothecin content of the anti-cancer plant Nothapodytes nimmoniana from shoot tip and nodal stem explants derived from in vitro seedlings was investigated. Multiple shoots induction from shoot tip explants was much greater than that from stem explants. The greatest shoots induced from the shoot tip and nodal stem explants was achieved on MS with 0.25~0.5 mg/L TDZ and 0.5 mg/L TDZ, forming over 20 shoots per explant in 10 week. Elongation of these multiple shoots was successful at a lower BA level (0.05~0.1mg/L) on MS medium. Individual shoots were rooted on MS supplemented with IBA or NAA. The 0.5mg/L IBA or NAA gave the highest percentage of rooting (83.3%) and root number (2.8~5.9 roots/shoot). Multiplication rates of shoot tip cultures derived from offspring of mother trees where grown at Green Island, Orchid Island, and Taitung provenances did not differ significantly. However, rooting rates of shoots were significant different among 3 provenances, which were ranging from 22.2 to 100%. Camptothecin (CPT) concentrations in leaves of seedlings and their mother trees in 3 provenances varied greatly. CPT concentrations in seedlings were much less than that in their mother trees, however, the positive relationship compare them to the mother plants was observed. Roots had high CPT level. In some cases, it contained CPT level 30 times greater than leaves.
