臺灣杉果膠甲基酯化酶基因之選殖及特性分析

Molecular Cloning and Characterization of Pectin Methylesterases from Taiwania Cryptomerioides

摘要


果膠甲基酯化酶(pectin methylesterases, PMEs)為催化細胞壁中之果膠成分進行去甲基酯化反應的一種酵素。它控制細胞壁的結構,參與花粉管的生長等之重要發育過程。經由臺灣杉Mago及Y14蛋白質釣取的基因庫中,選殖到果膠甲基酯化酶之基因,命名為TcPME2。TcPME2的轉譯區(coding region)共有1755個核苷酸,經由胺基酸序列比對發現TcPME2具有果膠甲基酯化抑制酶(PME inhibitor, PMEI)及果膠甲基酯化酶兩個功能區(domain),其蛋白質結構中包含多個平行β-平板和5個α-螺旋。另外,由反轉錄聚合鏈反應顯示,TcPME2在雌毬果及70年生的葉表現量較高,於雄球果、韌皮部、2個月小苗的根、莖及葉表現量較低,而木質部則無表現。

Pectin methylesterases (PMEs) are catalyse demthylesterification of pectins, which are major components of plant cell walls. They control the structure of the cell wall and are involve in important developmental process including pollen tube growth and so on. TcPME2 was obtained from yeast two-hybrid screening by TcMago-TcY14 heterodimer in Taiwania (Taiwania cryptomerioides Hayata). There were 1755 nucleotide in the coding region of TcPME2 that contains PME domain and PMEI domain. Protein structure of TcPME2 contained several β-sheets and five α-helixes. TcPME2 expressed higher in ovulate cone and 70-year-old leaf than microsporangiate cone, phloem, and stem, root and leaf of 2-month-seedling in Taiwania.
