紅毛杜鵑與內生菌Phialocephala sp.形成杜鵑類菌根

摘要


紅毛杜鵑(Rhododendron rubropilosum Hayata)為杜鵑科(Ericaceae)杜鵑屬(Rhododendron)之台灣原生樹種。本研究旨在探討紅毛杜鵑菌根之形態及微細構造,分離鑑定培養菌根菌,並接種菌根菌至無菌培育之紅毛杜鵑小苗,以瞭解其菌根特性。研究結果顯示,紅毛杜鵑之菌根有2種形態;其根部皮層細胞內有菌絲圈之微細構造,且所分離出菌根菌之菌落生長速率相當緩慢,具有杜鵑類菌根菌(ericoid mycorrhiza)之特徵;分離出之菌株以5.8S rDNA序列鑑定為Phialocephala sp.。接種Phialocephala sp.菌株至紅毛杜鵑小苗根部,發現接種苗生長良好且其根部皮層細胞內形成許多菌絲圈,證實紅毛杜鵑苗木可與內生菌Phialocephala sp.形成杜鵑類菌根。

Red-hairy azalea (Rhododendron rubropilosum Hayata), belonging to genus Rhododendron family Ericaceae, is an indigenous tree species in Taiwan. In this study, morphology and ultrastructure of mycorrhizal association of R. rubropilosum were observed. Mycorrhizal fungus was isolated, identified, and inoculated onto roots of R. rubropilosum seedlings in order to elucidate its mycorrhizal characteristics. The results revealed that the natural root association of R. rubropilosum could be distinguished into 2 morphological types, and there were hyphal coils in cortical cells. The isolated ericoid endophyte had slow hyphal growth rate, and was identified as Phialocephala sp. with 5.8S rDNA sequence. R. rubropilosum seedlings inoculated with Phialocephala sp. grew well and developed hyphal coils in cortical cells. These results demonstrated that R. rubropilosum seedlings could form ericoid mycorrhiza with Phialocephala sp.
