以牛乳中寡醣之生成評估褐藻酸β-半乳糖苷酶珠體之儲存方法

Evaluating Storage Methods for Alginate β-galactosidase Beads with Yield of Oligosaccharides in Milk

摘要


本研究旨在探討褐藻酸β-半乳糖苷酶珠體不同的儲存方式,對牛乳中寡醣生成之影響。用以製造褐藻酸β-半乳糖苷酶珠體之褐藻酸為2%之溶液。試驗分組方式為,將褐藻酸β-半乳糖苷酶珠體分別置於無菌水、0.1M CaCl2及0.3M CaCl2溶液中,並分別儲存於4℃及25℃,試驗期間為30日。主要測定的項目為β-半乳糖苷酶活性及寡醣生成量。由酵素活性及寡醣產量結果顯示,各處理組均隨著儲存期間之延長,而明顯降低酵素活性及寡醣之產量。由酵素活性變化中,顯示β-半乳糖苷酶珠體儲存於4℃之活性明顯高於25℃儲存者。綜合上述試驗結果顯示,β-半乳糖苷酶珠體置入0.3M CaCl2 溶液中,於4℃儲存,為生成寡醣時較適合之儲存方式。

This study investigated the effects of alginate β-galactosidase beads stored by different storage methods on yield of oligosaccharides in milk. Two percent Na-alginate solutions were employed to produce β-galactosidase beads. The beads were then stored in sterile distilled water, 0.1 M CaCl2 and 0.3 M CaCl2 at 4 or 25℃ for 30 days. The results showed that enzyme activities and oligosaccharide contents of all treatments decreased with increase in storage period. The beads stored at 4℃ had higher enzyme activities than those stored at 25℃. Among different storage conditions, alginate β-galactosidase beads stored in 0.3 M CaCl2 at 4℃ showed the best yield of oligosaccharides in milk.
