液體覆蓋對難微體繁殖蝴蝶蘭品種短莖培植體器內芽體增殖的影響

Effect of Liquid Overlays on In-vitro Shoot Proliferation of Recalcitrant Micropropagation Phalaenopsis Shoot Explant

摘要


部分市場熱銷的蝴蝶蘭品種,組培苗需求量大,卻受限於微體繁殖芽體增殖倍率低與芽體生長緩慢,被歸類為難微體繁殖品種。液體覆蓋兼具固體培養與液體培養的優點,本研究探討液體覆蓋對難微體繁殖蝴蝶蘭器內芽體增殖的影響。相對於固體培養,蝴蝶蘭滿天紅(Phal. Queen Beer)採用第6週使用液體覆蓋,第8週時芽數可達3.4個且第12週可得4.7個,不僅芽數為處理間最多且顯著縮短獲得目標芽數達3個之培養週期。無論第6週液體覆蓋培養基添加量為8.5 mL或17 mL,滿天紅芽數在往後6週的表現皆優於固體培養基。然而,相同條件的液體覆蓋在蝴蝶蘭Phal. I-Hsin Weston未見促進芽體增殖,可進一步探究其最適化的培養條件。

Some popular Phalaenopsis cultivars in the market have a large demand for tissue culture seedlings, but are limited by the low shoot proliferation rate and slow growth of shoot, so they are classified as recalcitrant micropropagation cultivars. Liquid overlay has the advantages of both solid medium and liquid medium. This study explored the effect of liquid overlay on in vitro shoot proliferation of recalcitrant micropropagation phalaenopsis. Compared with solid culture of Phal. Queen Beer, when the liquid overlay was added to the solid medium in the 6^(th) week, the number of shoots could reach 3.4 shoots per explant in the 8^(th) week, and 4.7 shoots per explant in the 12^(th) week, not only was the most among the treatments, but also was significantly shorten the culture period for obtaining the target shoot number up to 3. Regardless of whether the addition of liquid medium was 8.5 mL or 17 mL in the 6^(th) week, the number of shoots of Phal. Queen Beer was better than that of solid medium in the next 6 weeks. However, the liquid overlay couldn't promote the shoot proliferation of Phal. I-Hsin Weston under the same conditions, and its optimal culture conditions can be further explored.
