藥物誘生鼠瘤組織中巨分子化合物生合成之研究

Studies on the Macromolecule Synthesis Activities in Chemical Induced Rat Tumor Tissue

摘要


本文係報告鼠皮下瘤組織與正常皮下組織中,巨分子化合物(macromolecules)生合成活性(biosynthetic activity)之差異。該瘤組織係經黃麴毒素B1及四氯化碳處理Sprague-Dawley大白鼠皮下組織而獲得。上述兩組織中內源巨分子化合物合成酶之活性,係以離體培養(cultured in vitro)法測定之。結果顯示,經六十分鐘培養後,瘤組織之蛋白質生合成活性為正常者之十六點四倍。至於核糖核酸及去氧核糖核酸之生合成活性則分別為正常者之三點六倍及二十二倍。再者,上述兩組動物先以放射性尿核苷活體處理(treated in vivo)後,以層析法分離該兩組織之3'-聚腺核苷酸-核糖核酸(3'-poly A rich ribonucleic acid),結果顯示該瘤組織所含3'-聚腺核苷酸-核糖核酸之放射性比活性為正常者之二點二倍。若比較上述巨分子化合物生成活性之比值,則以去氧核糖核酸於瘤組織和正常組織中活性之比值為最高,這可視為瘤組織重要性質之一,表示瘤細胞核中基因複製之活性遠比正常者為高。

The differences of the endogeneous macromolecule synthesis enzyme activities between the subcutaneous tumor tissue and the control tissue had been studied in vitro. The results indicated that the protein biosynthetic activity in tumor tissue was 16.4 times higher than that in the normal tissue. The ribonucleic acid and deoxyribonucleic acid biosynthetic activities were 3.6 and 22 times higher than that in the normal tissue respectively. Furthermore, when the two groups animals were pretreated in vivo with 3H-uridine, the 3'-poly A rich RNA isolated showed that the specific radioactivity of poly A rich RNA from tumor was 2.2 titmes higher than that from the normal tissue. Comparing the ratios of these macromolecule synthesis activities in these tissues, DNA is the highest one. This is an important character of the tumor, indicating the DNA replication activity in the tumor is much higher that in the normal subcutaneous tissue.
