The Inhibitory Efficacy of Garlic Fractions on the Experimental Oral Cancer of Hamster

各種大蒜萃出物抑制倉鼠口腔癌發生之效能

摘要


大蒜抑制癌發生的效能,曾在本研究室中經過體內及體外系統的多項實驗獲得證實,但為求探討其作用的主要成分,及使用之安全性,本實驗即運用大蒜之五種萃出物在口腔癌的動物模式中,測定各萃出物的抑癌效能。使用125隻倉鼠,分別餵食含有5%或1%的五種大蒜萃出物乏飼料七週,並於第二週起,以0.5%DMBA溶液每週三次的塗抹於動物之左頰囊袋十週,以誘導生癌。另以對照組動物分別測定大蒜五種萃出物、DMBA、及礦物油之致癌性及對動物體能之影響,以供比較。實驗中每兩週記錄動物之體重、頰囊黏膜中血管之變化、和腫瘤形成之狀況,於第26週犧牲動物,立刻檢驗血液生化變化(SMA)、頰囊黏膜細胞中DNA合成量、和GGT酵素之動態,並將動物解剖,收集各器官之變化及腫瘤,做組織病理切片檢查。比較上述各檢查之結果後發現大蒜之C萃出物具有最佳的抑制口腔癌能力,且其5%濃度作用效果較佳。此C萃出物中的主要成分、作用機轉、對全身性癌症,如胃癌之抑制效果、及與其他大蒜精製化學成品效力之比較等,均為日後繼續深入鑽研之有趣課題。全部資料將對推廣「大蒜抑癌」之理想有莫大價值。

The inhibitory efficacy of garlic fractions on the experimental oral carcinogenesis of hamster was studied on 125 Syrian hamsters in order to explore the responsible major cancer inhibition components in garlic. Five fractions of garlic were prepared by different solvents extraction. The animals were divided into 10 experimental groups and 13 control groups to test the anticarcinogenicity of each fraction (oral administration of fractions followed by DMBA pouch painting) and the intrinsic carcinogenicity of the materials (garIic fractions, mineral oil and DMBA) respectively. The examinations of body weight, tumorigenesis, and the architecture of blood vessels of the pouches were performed biweekly before sacrifice. Right after sacrifice, the blood chemistry (SMA), DNA synthesis and GGTase activity of the pouch mucosa, and the autopsy of animals including histologic microscopy were carried out to obtain the necessary data as the criteria to judge the inhibitory efficacy of garIic fractions. Fraction C was found to possess the highest efficacy. SimiIar investigations will be taken place to further evaluate the cancer-inhibitory efficacy of garic fractions on the experimental gastric carcinogenesis before deeper exploration of the most efficient constituents of garlic in cancer prevention.
