Morphology and Growth Kinetics of Human Pterygium Fibroblast in Primary Culture

Background: To establish primary culture and investigate the characteristics of human pterygium fibroblast in vitro. Methods: The morphology of fibroblast was studied with Giemsa stain, scanning electron microscopic (EM), transmission EM and immunostaining of cytoskeleton. Growth kinetic studies were evaluated by growth curve, labeling index, cell cycle, doubling time, saturation density, plating efficiency and anchorage independent test. Results: We successfully establish a primary culture of pterygium fibroblasts in vitro. The fibroblast revealed elastodysplasia and dystrophy. The doubling time was 20 h and saturation density was 8 x 10^2 cell per cm^2. The plating efficiency was 36-43%, which was higher than that of normal cells (28-34%). Conclusions: Abnormal morphology and growth behavior of pterygium fibroblasts may explain the progressive nature of clinical pattern. This model could be used for further studies on pathogenesis, prevention and treatment of pterygium.
