Apoptosis and Graft Rejection in Rat Composite Tissue Allotransplantation

Apoptotis is thought to play a part in organ or tissue transplantation rejection. The cellular DNA of apoptotic cells is frafmented into pieces due the action of endogeous and can be detected by terminal deoxynu-cleotidyl transferase-mediated dUTP [LD1] nick-end labeling (TUNEL) staining. We studied apoptosis events in a rat model of limb allotransplantation. The number of TUNEL-positive cells on day 1 following transplantation increased in the allograft compared with the isograft, reaching a peak level on day 5. The TUNEL-positive cells were almost mvocytes [LD2] on day 1 and infiltrated mononuclear cells on day 5. The apoptosis events observed in this study revealed that apoptosis might play a role in acute rejection of rat limb transplantation.
