Hypolipidemic Effects of Inulin and Synthetic Oligofructose in Balb/c Mice

菊糖及生技果寡醣對小鼠脂質代謝之影響

摘要


本實驗主旨在探討菊糖及生技果寡醣對Balb/c小鼠之降血脂功效,以及這些作用是否與糞便脂質排出以及糞便短鏈脂肪酸濃度有關。七週齡小鼠隨機分為三組,各餵飼含有5%纖維素、菊糖或果寡醣之AIN-93G飼料三週,並於第18-20天收集糞便以分析中性固醇、膽酸、粗脂以及短鏈脂肪酸。實驗結束後,收集血液及肝臟以分析膽固醇、三酸甘油脂以及磷脂質。結果顯示,與纖維素相較,菊糖或果寡醣顯著降低血膽固醇濃度以及三酸甘油脂濃度。各組之肝臟膽固醇及磷脂質含量無顯著差異,但是果寡醣顯著降低肝臟三酸甘油脂含量。與纖維素相較,菊糖使小鼠每日糞便中性固醇排出量提高約1.5倍,而果寡醣則提高約0.6倍;菊糖及果寡醣各提高小鼠每日糞便膽酸排出量約一倍;菊糖及果寡醣亦顯著降低外表脂質吸收率。在糞便短鏈脂肪酸濃度方面,各組之乙酸濃度無顯著差異,然而菊糖及果寡醣皆提高總短鏈脂肪酸濃度、丙酸濃度以及丙酸之比例。因此本實驗建議菊糖及生技果寡醣之降血脂效應與糞便中脂質之排出以及其發酵產物有關。

The aim of this study was to investigate the modulatory effects of inulin and synthetic oligofructose (OFS) on blood lipid levels, and whether these effects are associated with fecal excretion of lipids and fecal short-chain fatty acids (SCFAs) in Balb/c mice. Seven-week-old mice were fed modified AIN-93 diets containing 5% cellulose, inulin, or OFS for 3 weeks. The cholesterol, triacylglycerol, and phospholipids levels in the plasma and liver were determined. Feces collected during days 18~20 was analyzed for neutral steroid, bile acid, crude fat, and SCFAs. Both inulin and OFS significantly decreased the plasma cholesterol and triacylglycerol levels as compared with cellulose. Most of the hepatic lipid contents were similar across groups, except that OFS significantly reduced the hepatic triacylglycerol content. Inulin elevated the daily neutral steroid excretion by ~1.5-fold while OFS slightly increased the neutral steroid excretion by 0.6-fold, as compared with cellulose. Inulin and OFS similarly caused a 1-fold increases in fecal bile acid excretion. Furthermore, the apparent fat absorption in mice fed inulin and OFS diets were similar, 78.7%(5.3% and 77.6% (4.7%, respectively, which were significantly lower than that of mice fed the cellulose diet, 88.7% (5.9%. The fecal acetate concentration did not significantly differ among groups. However, inulin and OFS significantly promoted the concentration and proportion of propionate in the feces. In conclusion, increased fecal excretion of steroids, reduced fat absorption, and increased production of propionate may mediate the hypolipidemic effects of inulin and OFS in Balb/c mice.
