膳食油脂的種類與量對卵蛋白致敏BALB/c小鼠過敏免疫反應的影響

Effects of Different Dietary Oils and Contents on Immune Responses of Ovalbumin Sensitized BALB/c Mice

摘要


為探討膳食油脂的攝取量與形態,是否對目前日益增加的過敏免疫疾病有所影響,本實驗以卵蛋白(OVA)致敏BALB/c小鼠的動物模式,分別餵食低劑量或高劑量的不同種類的油脂,探討對過敏免疫反應的影響。實驗步驟為將十週大小鼠隨機分為6組,分別餵食飼料含5%低劑量的奶油(LB組)、芥花油(LC組)、或黃豆油(LS組)、15%高劑量的奶油(HB組)、芥花油(HC組)、或黃豆油(HS組)五週後,進行隔週以OVA腹腔注射致敏共三次,並以PBS組以PBS取代OVA作為致敏對照組,隔週進行吸入性OVA致敏後犧牲。結果顯示膳食油脂對OVA致敏小鼠的體重、攝食效應和組識重等無顯著影響,對過敏免疫反應的影響,高油飲食降低致敏小鼠脾臟細胞IFNγ與IL-2分泌,及血清○VA特異性IgG1與IgG(下标 2a)生成;油脂種類則顯著影響肝臟脂肪酸組成、脾臟細胞IL-2和IFNγ分泌、肝臟外因性前列腺素E2(exo-PGE2)及尿液bicyclo-PGE2含量。芥花油組有較高的脾臟細胞分泌PGE2能力,高量黃豆油與芥花油分別含有高量18:2和18:1脂肪酸,肝臟exo-PGE2及尿液bicyclo-PGE2含量顯著增加,脾臟細胞IL-2分泌顯著降低。奶油富含14:0和16:0飽和脂肪酸,肝臟exo-PGE2及尿液bicyclo-PGE2含量顯著最低,高劑量奶油顯著抑制肺沖洗液嗜伊紅性白血球聚集,但低劑量奶油反而顯著促進,故奶油攝食量對過敏性氣喘的影響,仍需進一步的探討。

To investigate whether amount and type of dietary fat may exert effect on allergic immune responses, 10-week-old female BALB/c were divided into six groups and fed with AIN76 diet containing either 5% butter (LB), 5% canola oil (LC), 5% soybean oil (LS), 15% butter (HB), 15% canola oil (HC), or 15% soybean oil (HS). After 5 weeks' experimental diet, mice were sensitized intra-peritoneally and challenged with ovalbumin (OVA) every other week for four times. The results showed that different dietary fat did not affect body weight, tissue weight and feed efficiency. High fat that the content of dietary oil affected the intake, cytokine sproduction of splenocytes (IL-2, IL-6, IFNγ), and the titer of anti-OVA antibodies (IgE, IgG1, IgG(subscript 2a)) in the serum of OVA-sensitized mice. Different kind of dietary oil affected the hepatic fatty acid composition, PGE2 and cytokine production of splenocytes (IL-2, IL-6, IFNγ), exo-PGE2, bicyclo-PGE2 of urine, and the percent of eosinophil in bronchoalveolar lavage fluids (BALF). 15% high fat diet had the potential of increasing PGE2 production of splenocytes, and could also down-regulate the production of Th1-type cytokine such as IL-2 of splenocytes. Oil contain low 18:2 (butter) could up-regulate Th2-type cytokine, IL-6, production of splenocytes, but it didn't increase the production of exo-PGE2 and bicyclo-PGE2 in body. On the other hand, high content of butter could significantly inhibit the accumulation of eosinophil in BALF.
