Cellulase Production by Xylaria Regalis, a Wood Grown Ascomycete

木生子囊菌Xylaria regalis產生織維素分解酵素之研究

摘要


本研究旨在探討分離自本省之一株木生子囊菌Xylaria ragalis產生織維分解酵素之能力。將X. regalis菌株培養於28℃,二星期後,其endo-β-glucanase、cellobioriydrolase及B-glucosidase三種織維素分解酵素之最高產量依次為65.3mU/ml、21.7mU/ml及55.6mU/ml,比較X. regalis及Trichodema reesei ATCC26921 (=QM9414,為已知之cellulase高生產菌株)二者產生織維素分解酵素之能力,發覺前者之β-glucosidase產量較後者高,但endo-β-glucanase及cellobiohydrolase之產生量則低於後者。X. regalis所產之endo-β-glucanase、cellobiohydrolase及β-glucosidase三種織維素分解酵素之最適作用酸鹼值依次為4.0、5.0及5.0;最適作用溫度則為40、40及50℃。

Production of cellulolytic enzymes by the wood-grown ascomycete Xylaria regalis isolated in Taiwan was studied. The highest levels of endo-β-glucanase, cellobiohydrolase, and β-glucosidase produced by X. regalis were 65.3 mU/ml, 21.7 mU/ml, and 55.6 mU/ml, respectively, when cultivated at 28℃ for two weeks. X. regalis produced more β-glucosidase but less endo-β-glucanase and cellobiohydrolase in activity than Trichoderma reesei ATCC 26921 (=QM 9414). When culture filtrate was used as a crude enzyme solution, the optimum pHs for endo-β-glucanase, cellobiohydrolase, and β-glucosidase activities were 4.0, 5.0, and 5.0, the optimum temperatures being 40, 40, and 50℃, respectively.
