A Preliminary Screen of Antibiotics Produced by Thermophilic Fungi Isolated from Taiwan

臺灣嗜熱性真菌生產抗生物質篩選之初探

摘要


由未探索過的生物資源中篩選新的生物活性物質是一種相當實際的篩選策略。臺灣嗜熱性真菌採集調查結果發現了不少新種及新紀錄種。近千株分離菌株經菌種分類篩選、抗菌活性篩選及化學結構篩選三個階段處理後,發現其中有新抗菌活性物質,其產生有溫度依賴性。Malbranchea 90354產生琨類(quinone)抗生物質,其中一種是別的微生物中未曾發現過的;Myceliophthora90451産生5種chromine類抗生物質,生產物質及其醯化物經紫外光、質譜以及氫及碳13核磁共振圖譜分析,證實其中一種為真菌代謝物chaetochromine,其他物質分別為benzochromin類及naphthoauinone類,其化學結構尚待證實,有二種代謝物質生物活性測試結果顯示具抗菌及抗癌活性。

Screen of new biologic metabolites from newly found organisms is used as a practical strategy. In Taiwan, during comprehensive investigations of thermophilic fungi, new species and new record strains were recognized. After a preliminary screen, very high ratio was shown in producing bactericidal substances. Several possibly new antibiotics were suggested to be produced by these precious fun- gal strains. Among strains used, Malbranchea was proved to produce quinone antibiotics, and Myceliophihora produce several chromins. Temperature dependent production was noticed. The unstable properties and conjugation tendency makes it complicate for further study. A novel quinone with an acetyl-ethyl substitute was identified from its acetylation derivatives. A known compound chaetochromin was found by Myceliophthora from acetylation derivatives also. Another three chromine compounds were elucidated as chaetochromin and benzochromin according to the MS and NMR spectrum.
