Isolation, Purification, and Characterization of the PR-amide Synthetase from Penicillium roqueforti

藍酪黴菌PR-amide合成酵素之分離、純化及其生化特性之探討

摘要


PR-acid舆PR-amide是藍酪黴菌所產生之次級代謝產物,由PR-amide合成酵素將PR-acid轉化成PR-amide。我們培養19天的菌絲磨出液,經ultra filtration濃縮 、80%硫酸銨鹽析,疏水性層析管柱(Phenyl)以及HPLC膠體層析管柱純化,得到最終產物活性能提高5.8倍,回收率為5.8%。經由SDS-膠體電泳分析得知此酵素分子量為66 kDa;此外,此酵素最適反應pH值為5.0,最適反應溫度為40℃。

PR toxin, PR-acid, and PR-amide are the secondary metabolites of Penicillium roqueforti. The chemical structures of these three compounds are closely related to each other. In an effort to understand the degradative pathway of PR toxin, we found the PR-amide synthetase. The enzyme involved in the conversion of PR-acid into PR-amide, was purified from the culture mycelium of Penicillium roqueforti ATCC48936. The enzyme has a molecular mass of approximately 66 kDa, and it is a monomer. The optimal pH for this enzyme is ca. 5.0, and the optimal temperature is 40℃.
