亞腐霉菌產生孢囊與游走子技術之建立

Stimulation on production of sporangia and zoospores in Pythiogeton species

針對台灣產9種供試亞腐霉菌(Pythiogeton abundans、P. microzoosporum、P. oblongilobum、P. paucisporum、P. proliferatum、P. puliense、P. ramosum、P. uniforme 及P. zizaniae)進行孢囊及游走子產生之測試。試驗結果顯示固態培養基並不適合孢囊之產生;但若將供試菌培養於盛有10 %離心蔬菜汁(10 % CVB)之培養皿中培養2-3天以產生菌絲團,再以未滅菌之茭白田水置換並於24 °C培養2天,則所有的供試菌皆可產生大量孢囊,其孢囊產量高於以無菌水、滅菌茭白田水或過濾膜過濾茭白田水等置換培養,但縱使有孢囊產生,除了少數菌株有少量的孢囊釋放游走子外,大部分的菌株並無游走子之釋放。實驗發現,需將上述培養皿內之茭白田水倒棄一部分,在此培養皿內餘留部分之茭白田水,然後將蓋玻片覆蓋於產有孢囊之菌絲團上數小時後,則可促進所有菌株之游走子大量產生,而所有供試菌的孢囊產生與游走子形成之最適溫為24 °C或28 °C。

Methods were developed for stimulating Pythiogeton species obtained in Taiwan to produce sporangia and zoospores. The species tested in this study were P. abundans, P. microzoosporum, P. oblongilobum, P. paucisporum, P. proliferatum, P. puliense, P. ramosum, P. uniforme and P. zizaniae. For the production of sporangia, liquid medium, 10% clarified V8 juice broth (10% CVB), were applied. After mycelial mats had been well-developed in the broth, the broth was replaced experimentally by sterile distilled water and treated/untreated field water; the mycelia were then incubated for two more days. Among these tests, sterile distilled water was not suitable for sporangia production, while unsterilized field water could stimulate sporangial production most prolifically. Although abundant sporangia had been induced, there were barely zoospores released from the sporangia. To induce the massive releasing of zoospores, the mycelial mats with mature sporangia submerged in a small amount of field water in Petri dishes needed to be covered with cover-slips for hours. The optimal temperatures for production of sporangia and zoospores of all the tested Pythiogeton isolates were 24°C or 28°C.
