Using a Novel Molecular Diagnostic Technique for Detection of Emerging Unknown Pathogens

摘要


新興感染症已成爲全球健康的重要議題,其中新興病毒引起的感染症最爲顯著。本研究中我們建立了Virus Discovery cDNA-AFLP (VIDISCA)分子鑑定平台用於偵測疑似病毒感染的臨床檢體或樣本。在上呼吸道感染檢體中,可偵測到Human rhinovirus 43、Staphylococcus spp.、Micrococcus luteus以及Mycoplasma fermentans。在慢性阻塞性肺病檢體中,找到Pseudomonas spp.、Ascomycota sp.與Actinomycetospora sp.。此分子鑑定平台的建立不僅能偵測樣本中的未知致病原,提供相關疾病診斷與治療的有利資訊,更可以對未來新興病毒的檢測技術奠定良好的基礎。

Objective: Emerging infectious diseases have become an important issue for global health and newly emerging viruses are the major threat to human health among the infectious diseases. In this study, we established a novel molecular diagnostic platform, Virus Discovery cDNA-AFLP (VIDISCA), to detect clinical specimens or samples derived from patients suspected of viral infections. Through this approach, human rhinovirus 43, Staphylococcus spp., Micrococcus luteus and Mycoplasma fermentans were found in samples from patients with upper respiratory tract infections. In addition, Pseudomonas spp., Ascomycota sp. and Actinomycetospora sp. were identified from specimens of patients with chronic obstructive pulmonary diseases. Establishment of VIDISCA technique platform can identify the unknown pathogens in specific infectious diseases to provide valuable information for clinical diagnosis and treatment. Furthermore, it can serve as a solid foundation for detection of newly emerging viruses in the coming future.
