高尾山蟲草無性世代(Paecilomyces tenuipes)發酵產物在抗氧化及促進吞噬能力之初步研究

A Preliminary Study on the Anti-oxidant and Phagocytosis from the Fermentation of Anamorph Paecilomyces tenuipe

摘要


蟲草屬真菌(Cordyceps spp.)是一群藥用真菌,具有廣泛的生物及藥理活性,在許多藥理學研究中可發現,蟲草屬真菌具有抗腫瘤、免疫調節、抗氧化及保肝等作用。本篇研究題材為高尾山蟲草無性世代(Paecilomyces tenuipes),探討其發酵產物在抗氧化及提升老鼠巨噬細胞(RAW 264.7)吞噬能力。實驗結果發現,P. tenuipes發酵後產物具提升巨噬細胞吞噬能力,以菌絲體熱水萃取物(1mg/mL)為最佳。熱水萃取菌絲體具有刺激老鼠巨噬細胞產生一氧化氮(NO)能力,且具有劑量遞增性效果。在抗氧化能力方面,發酵液具有不錯清除DPPH能力。再進一步使用流式細胞儀來偵測CHO-K1細胞氧化程度。結果顯示,高尾山蟲草發酵產物可降低H2O2對CHO-K1細胞內產生的氧化傷害。希望這些發現可對高尾山蟲草研究有所助益。

The genus Cordyceps is a group of precious medicinal mushrooms which possess a wide of biological and pharmacological activities. The medicinal effects, such as anti-tumor, immunomodulation, antioxidant activity and hepatoprotective effect, had been studied in past years. This study on the anti-oxidant and Phagocytosis from the fermentation of anamorph Paecilomyces tenuipe. The hot crude extract of mycelia could significantly enhance phagocytosis activity on RAW 264.7 (p<0.05). Meanwhile, enhancing nitric oxide activities on macrophage cellsin adose-dependent evaluated by Griess method. In the anti-oxidized aspect, the fermentation filtrate had also shown a great activity by scavenging the DPPH. The antioxidant activity was assayed by Flow cytometry, and the result showed that the fermentation filtrate had a great antioxidant activity. These results suggest that Paecilomyces tenuipe indeed possessed some biology activities. Hopefully, these findings are useful to further studies on bioactivity of Paecilomyces tenuipe.
