摘要


癌症的死亡大都是由於對轉移癌細胞的治療失敗所致,因而研究癌症轉移機制已成為研究癌症的終極目標,在此review我們簡單討論,就我們現今所知癌細胞經由(1)活化epithelial mesenchymal transition(EMT),(2)extracellular matrix(ECM)的remodeling及(3)癌細胞由原位轉移到特定的器官等三種機制完成癌細胞轉移。由我們及其他研究同儕的研究結果顯示,屬於ETS群的轉錄因子在活化EMT及ECM remodeling扮演重要的角色,因此研究在這三種轉移機制的調控,將有助於了解癌細胞如何由良性腫瘤變成轉移性癌細胞的過程中,基因及epigenetic的變化。

Most cancer deaths are attributed to the failure of response to treatment of migrating tumor cells. Thus investigation of the mecha-nisms of tumor invasion and metastasis has become the last frontier of cancer research. Here we briefly discuss how benign cancer cells, through well orchestrated mechanisms of (a) activation of epithelial mesenchymal transition (EMT), (b) remodeling of extra-cellular matrix (ECM), and (c) migration of cancer cells to distant specific sites, turn into metastatic cancer cells. From the experi-mental observations by us and others we know that members of the ETS transcription factor family play important roles in activating genes encoding proteins responsible for EMT and ECM remodeling. Thus studies into the regulatory mechanisms of these three events will help trace the genetic and epigenetic changes that turn a benign cancer cell into a malignant migrating one.
