Molecular Studies of an Acrosomal Antigen Recognized by a Monoclonal Antibody and Its Application in Monitoring Sperm Acrosome Reaction

A conserved sperm acrosome antigen which reacts specifically with HS-11 monoclonal antibody was purified by immunoaffinity chromatography. The isolated antigen from mouse testis extract designated as MSA-11 was shown to belong to a group o fproteins with molecular mass ranging from 30 to 50 KD on SDS-PAGE. By using two-dimensional gel eletrophoresis, MSA-11 was resolved into 7 distinct proteins with isoelecric points ranging from 4.4 to 4.7. In the native state, MSA-11 was shown to be high molecular weight aggreagates as judged from Sephacryl S-300 gel filtration chromatography. Similar to HS-11 monoclonal antibody, the corresponding mouse or rabbit antisera also reacted with the acrosome of human or mouse sperm. When the monoclonal antibody was incubated with live human sperm in the capacitation medium, the percentage of methanol-fixed sperm stained with indirect immunofluorescence was found to decrease significantly with the induction of sperm acrosome reacton following the treatment with calcium ionophore A23187. The results of this study suggest that HS-11 reacts with sperm antigen which is exposed after sperm capacitaion and is shed after acrosome reaction.
