Adenovirus-Mediated Gene Therapy Using IL-12 and Angiostatin Genes for Murine Lung Cancer

Lung cancer is the leading cause of cancer death in the world, and also in Taiwan, In the last few decades, the prognosis of lung cancer patients has little altered with conventional therapies. Gene therapy, therefore, may be a promising alternative in the treatment of lung cancer. Methods: In this study we evaluate the effect of adenoviral vectors encoding IL-12 (ad/IL-12) or angiostatin used singly or in combination with cisplatin treatment or tumor resection to treat Lewis lung cancer in mice. Results: Our results show that Ad/IL-12 or Ad/AST alone at low doses exerted miner antitumor activities. However, the combination of cisplatin and Ad/IL-12 treatment reduced both the incidence and size of tumors if chemotherapy was given prior to, rather than after, the Ad/IL-12 treatment. Moreover, in the postoperative gene therapy model mimicking a status of tumor-relapse after surgery, the growth of reinoculated tumor cells was significantly suppressed in mice treated with Ad/IL-12 compared with those treated with control viruses. Animal experiments using different therapeutic regimes are still underway. Conclusions: Adenovirus-mediated IL-12 gene therapy, in combination with chemotherapy or surgery, may be promising strategies for the treatment of lung cancer.
