Evaluation of Chinese Herbs That Affect the Cell-Mediated Immunity (Ⅰ)

三十種中草藥對於細胞性免疫反應影響之評估(Ⅰ)

摘要


中草藥在我國已被臨床中醫師及民間廣泛運用,但目前仍缺乏有效科學證據以證實其療效,本篇論文即針對民間常用之消炎、止痛、清熱、解毒及抗腫瘤等三十種中草藥當材料,以細胞性免疫反應爲研究模式,藉以瞭解這些中草藥療效與細胞性免疫反應間的關係。實驗中,我們以甲醇或水進行天然物的粹取,所得之粗粹取物與人類單核細胞共同培養,經過三天以3H-thymidine標識去氧核醣核酸合成,由此可知中草藥之粗粹取物對於人類單核細胞生長之影響。結果顯示,在三十種具有清熱、解毒、消炎、陣痛之中草藥中,其中十六種對於人類單核細胞生長具有抑制作用,部份中草藥對於人類單核細胞生長之抑制作用是透過抑制第二個白質分泌而影響人類單核細胞增殖,其它中草藥則透過未知機制抑制細胞生長。這些實驗結果符合中草藥之作用,未來我們將純化其中之天然物,以探討其作用機轉。

Thirty Chinese herbs commonly used as antiinflammatory drugs in traditional Chinese medicine were selected for immunopharmacological activity test. Human mononuclear cells (HMNC) were used as target cells. Methanolic or water extracts of the Chinese herbs were added into HMNC cultures. Subsequent, cell proliferation were determined by 3H-thymidine uptake. The results indicated that 16 out of 30 methanolic crude extracts inhibited the HMNC proliferation. Only 3 out of 30 water extracts of Chinese herbs (MLS-024, MLS-027 and MLS-030) contained inhibitory agents that suppressed HMNC proliferation. The inhibitory mechanisms may involve the blocking interleukin-2 (IL-2) production. Since adding IL-2 to HMNC cultures, restore cell proliferation. Plans are underway for the isolation of pure compounds from these Chinese herbs and the elucidation of their mechanisms of action.
