絞股藍抽取物對兩步驟洗滌法人類精子活動力的體外效應評估

Effects of Gynostemma Pentophyllum Makino Extracts on Human Sperm Motility with Two Step Washing Method in Vitro

摘要


本研究探討具抗癌、抗衰老及強壯固精等多種作用的絞股藍抽取物,對體外人類精子活動力的效應。精液樣本依世界衛生組織之精液分析標準,分爲正常與異常兩組各17例,正常組的精子濃度每西西大於兩千萬隻,活動力大於50%;異常組的精子濃度每西西大於兩千萬隻,活動力介於20~40%。兩組各分HTF對照組,與濃度0.01mg/ml和1.0mg/ml之絞股藍溶液實驗組,實驗時先將精液樣本以兩步驟洗滌法處理,然後用美國漢彌爾頓電腦輔助精液分析儀(HTM-2030)分析HTF培養液和絞股藍抽取物在第一小時與第二小時,對精子「活動力」與「前進活動力」的影響。 絞股藍溶液對兩步驟洗滌法處理過之正常組與異常組精子「活動力」、「前進活動力」的刺激效應,在濃度效應上並無顯著意義;然而在時間效應上,正常組HTF對照組和絞股藍溶液1.0mg/ml,對精子「活動力」有刺激效應;異常組HTF和絞股藍溶液1.0mg/ml和0.01mg/ml,對精子「活動力」均有刺激效應。就精子「前進活動力」的刺激效應而言,正常組HTF有統計學意義,而絞股籃溶液則無統計學意義;異常組HTF和絞股藍溶液均有顯著意義。整體而言,HTF對正常組和異常組精子的「活動力」、「前進活動力」均有刺激作用,而絞股藍溶液對精子「活動力」、「前途活動力」的刺激作用,顯然異常組優於正常組。從中醫學的觀點而言,絞股藍對精子活動力差的人有強精作用,且與時間效應有關;但對正常人,絞股藍不一定有強精作用。

Given that Gynostemma Pentophyllum Makino (GPM) possesses multiple functions of anti-cancer, anti-aging, strengthening and securing essence in the traditional Chinese medicine, we devised the present study to assess the effects of GPM extracts on human sperm motility in vitro. The study was based on 34 eligible semen samples performed by the computer assisted semen analyzer according to the semen criteria in the WHO. Seventeen samples with a concentration over 20 million per milliliter and a motility over 50% placed in the normal group, and 17 samples with a motility between 20% and 40% placed in the anomalous group. Each sample was prepared by a two-step washing method, before observing the effect of HTE 0.01 mg/ml and 1.0 mg/ml GPM solution on sperm motility as well as progression at the first hour and the second hour. The results show that the concentration effects of GPM solution on sperm motility and forward progression were not significant in either the normal or the anomalous group. The facilitation of HTF media on sperm motility and forward progression was significant in both groups on the time effects. In the anomalous group, the facilitation of GPM solution on sperm motility and forward progression was superior to the normal group. In terms of Chinese medial theory, Gynostemma Pentophyllum extracts strengthen essence in low levels of sperm motility but not necessarily in normal semen.
