當歸、丹參萃取物對抑制癌細胞之影響

Cytotoxic Effects of Angelica acutiloba and Salvia miltiorrhiza Extracts on Human Cancer Cell Lines

摘要


以花蓮地區優良農業操作(GAP)栽培體系所生產的當歸與丹參為材料,利用不同方式萃取其有效成分,以進行抑制癌細胞增生之研究,結果證實當歸與丹參萃取物均能有效抑制體外癌細胞生長。當歸與丹參以80℃乙醇處理可萃取出最高有效成分含量,且可抑制MCF-7、MSG2 及A549 等癌細胞株的生長。當歸、丹參與順鉑相互之間的聯用處理組合,對HepG2肝癌細胞皆有抑制效果;在457 μg.ml^(-1)當歸萃取物與40 μg.ml^(-1)丹參萃取物的處理濃度組合時,細胞的存活率為20%;當歸萃取物與順鉑藥物聯用的處理組合,在457 μg.ml^(-1)當歸萃取物與5 μg.ml^(-1)順鉑處理濃度時,細胞的存活率為10.9%;丹參萃取物與順鉑藥物聯用的處理組合,在40 μg.ml^(-1)丹參萃取物與5 μg.ml^(-1)順鉑處理濃度時,細胞的存活率為50%。當歸、丹參萃取物與順鉑藥物三者聯用時,在457 μg.ml^(-1)當歸萃取物、40 μg.ml^(-1)丹參萃取物及5 μg.ml^(-1)順鉑處理濃度時,細胞存活率為31%。試驗結果顯示,當歸與丹參的有效成分與順鉑化療藥物聯用,有助於抑制HepG2肝癌細胞之增生,在未來抗癌與藥物動力學之臨床研究,可提供抗癌配方之應用,期能減少因化療藥物使用所產生的副作用,對未來開發保健產品及天然植物藥之應用上具有發展潛力。

Various combinations of Angelica acutiloba and Salvia miltiorrhiza extracts produced by GAP cultivation system in Hualien were confirmed in this study to inhibit the cancer cells proliferation in vitro. Those extracts with ethanol at 80℃ produce the highest content of active ingredients and inhibit the growth of MCF-7, MSG2 and A549 cell lines, and when combined with cisplatin can inhibit the proliferation of HepG2 cell line. Treated with combined extracts of 457 μg•ml^(-1) Angelica and 40 μg•ml^(-1) Salvia extract, cancer cell survival rate is 20%. Treatment of 457 μg•ml^(-1) Angelica extract and 5 μg•ml^(-1) cisplatin, the cancer cell survival rate is 10.9%. The combined treatment of 40 μg•ml^(-1) Salvia extract and 5 μg•ml^(-1) cisplatin resulted in 50% cancer cell survival. Combined with all three, it resulted in 31% cancer cell survival. This study provides evidence that when combining active ingredients of Angelica acutiloba and Salvia miltiorrhiza with chemotherapeutic agent cisplatin, it inhibits HepG2 cell line that can be used in future anticancer and pharmacokinetic research, benefit the application of synergistic combination to reduce the side effects of chemotherapy drugs. There are potential applications in the development of functional food and herbal medicine in the future.
