Mitomycin-C對培養中的纖維母細胞之抑制效果

A Study of Inhibitory Effect of Mitomycin-C on Fibroblasts in Cell Culture

摘要


青光眼濾過手術常因引流通道處之鞏膜外瘢痕纖維組織形成,而使其喪失功能,造成手術失敗。瘢㾗纖維組織之形成,主因纖維母細胞之增殖所致。故手術中以Mitomycin-C來抑制纖維組織之增值,當可增加手術之成功率。本實驗以取自眼球筋膜組織(Tenon's capsule)之纖維母細胞作組織培養,再以之測定Mitomycin-C對細胞增殖之抑制效果;進而求得Mitomycin-C對Fibroblast之百分之五十致死量(LD50)為0.2mg/ml,百分之五十增殖抑制量(50% Effective dose, ED50)為9.55×10^(-3)mg/ml。再以二者之比率求得Mitomycin-C之治療指數(Therapeutic index=LD50/ED50)為20.9,以供臨床使用之參考。

An in vitro study was made of the inhibitory effect of mitomycin-C on proliferation of Tenon's capsule fibioblasts in a cell culture. The materials for the cell culture were taken from Tenon's capsule during a glaucoma operation. Plating efficiency counting was used for the evaluation of mitomycin-C inhibitory effect. The plated cells were treated with mitomycin-C in concentrations from 1-400 μg/ml for five minutes. The cells were then observed under a phase contrast microscope, a light microscope using trypan blue stain and Giemsa's stain, and a transmission electronic microscope. The LD50% was 0.2mg/ml and the ID50%, that is, the ED50% was 9.55×10^(-3) mg/ml. The therapeutic index was calculated as 20.9, In vitro mitomycin-C prominently inhibits fibloblasts from proliferating and has potential cell toxicity. After filtering operation for glaucoma, fibrous tissue proliferation often negates the filtering effect of surgery. The present study provides a support to our clinical observation that local application of mitomycin-C to the operation site on exposed subconjunctival, Tenon's capsule, and scleral tissue during the procedures inhibits the undesired excessive fibrous tissue proliferation of the conjunctival flap and enhances the IOP control effect of surgery.
