以掃描電子顯微鏡觀察角膜上皮細胞外傷後之再生

Scanning Electron Microscopy of Epithelial Wound Healing in the Rabbit Cornea

摘要


以百分之九十五濃度之乙醇將兎眼之角膜上皮細胞刮除,並分別在第1小時,24小時,45小時,72小時,96小時和一星期將角膜取下,以掃描電子顯微鏡觀察再生之上皮細胞形態之改變,發現經乙醇處理後之角膜其基底膜依然完整,在其邊緣可看到向傷口之偽足狀細胞突起,所生之上皮細胞呈條狀,細胞形狀不規則,表面不平滑,且其上之細胞絨毛極不明顯,但四天後細胞即呈現相當平滑與對稱,且其上佈滿絨毛;七天後即與對照組之角膜相似,開始出現即將剝落之上皮細胞。由本實驗發現在沒有合併症發生之角膜上皮創傷癒合是藉細胞本身之滑行增生等功能而快速且完整的癒合,因此在臨床上對類似之角膜上皮刮傷之治療,應著重於避免併發症之發生。

The rapid and complete repair of the corneal epithelium following ocular surgery or trauma is essential for the maintenance of normal visual acuity. In this report we treated the rabbit cornea with 95% ethyl alcohol to study the morphology change of the corneal epithelium during wound healing with scanning electron microscopy (SEM). The cornea treated with ethyl alcohol results in removing all of the corneal epithelial cells but leaved basement membrane intact. The corneal epithelial defect was complete healed within four days, but it took another two days for complete the morphology change and maturity. We found the early stage of the renewal epithelium showed band in shape, uneven surface and rare in microprojections, but with the time gone it changed to flat, polygonal shape, straight cell boundaries and the surface fulfill with microprojections. This study showed the corneal epithelium wound healing rapid and complete if there is no complication occurred.
