為了解糖尿病老鼠視網膜多巴胺第一型受器活性的變化,本篇以不同的方法包括(1)靜脈注射streptozotocin(2)靜脈注射Alloxan及(3)胰臟切除(pancretectong)來引致糖尿病,再比較其視網膜多巴胺第一型受體活性。結果顯示出糖尿病老鼠視網膜第一型受體活性的結合速率有明顯下降,其結果表示在糖尿病老鼠的視網膜在早期的糖尿病高血糖時其視網膜多巴胺第一型受體此有明顯變化。
Alterations of dopamine DA-1 receptor in retina, which may be associated with the development of diabetic retinopathy, were investigated using (1) intravenous injection of streptozotocin, (2) administration of alloxan, and (3) application of pancreatectomy induced hyperglycemic rats. The retina from these diabetic rats was isolated for study of receptor bindings using [3H] SCH 23390 as radioligands. Activity of dopamine DAI receptors was then compared with the control group at the 3rd date of expermintal diabetic rats. The binding affinity (Kd values) of DA-1 binding sites measured with [3H] SCH 23390 in rat retina decreased at the 3rd date of hyperglycemia. These results suggest that retinal dopamine DA-1 receptors were changed at the early stage of acute hyperglycemic rats. The Decrease of binding affinity is depent on hyperglycemia, but not the drug toxicity.
