Immunohistochemical Localization of bFGF in Developing Chick Embryo Neuroretina

鹽基性纖維芽細胞成長因子在雞胚神經網膜層的免疫組織化學定位

摘要


運用免疫組織化學反應及電子顯微鏡檢查,我們在發育中的雞胚神經網膜層中,定位出鹽基性纖維芽細胞成長因子(bFGF)。 在胚胎第七天時,bFGF可在神經節細胞、苗勒氏細胞及星狀細胞被定位出,其反應位置主要位於細胞質。在胚胎第十一和第十四天時,神經節細胞層仍然有最強的免疫反應;在神經纖維層中的神經節細胞和苗勒氏細胞樹突胞質也同時顯出強的反應;此外,在內神經顆粒層,有部分移行的神經節細胞、星狀細胞和寡樹突膠質細胞也呈現免疫反應。在胚胎孵化後第一天,神經節細胞層和感光體層的內節顯出極強的免疫反應;同時,內神經顆粒層和外神經顆粒層也顯示出中等度的染色反應。 由本實驗中,我們推論在雞胚神經網膜層發育期間,bFGF具有分化、移行和細胞分裂的功能。此外,由於bFGF被定位的時間和位置,與Transferrin轉移受體及高量的粒腺體等發生時間數量相當一致;所以,我們推論,除了一般所知的神經性發生、滋養功能之外,bFGF對於發生、發育期間的新陳代謝調節功能,也佔有一定的角色和重要性。

We localized the basic fibroblast growth factor (bFGF) on the developing chick neuroretina by using immunohistochemistry and electromicroscopy. At E7, bFGF could be detected on the ganglion cell layer (GCL) with a special location in cytoplasm of ganglion cell, müller cell and astrocyte. At E11 and E14, the immunoreactivity of GCL was the strongest. Besides, processes of ganglion cell and cytoplasm of müller cell both extended ongrowth to nerve fiber layer (NFL) and they also demonstrated strong reactivity. The immunostaining was also found in some displaced ganglion cell, astrocyte and oligodendrocyte in inner nuclear layer (INL). At P1, both GCL and inner segment of photoreceptor layer showed strong reactivity. The INL and outer nuclear layer (ONL) revealed moderate staining. From the study, we conclude that bFGF has the differentiative, migrative and mitogenic functions in the developing chick neuroretina. Furthermore, the appearing time and location of bFGF was parallel with the transferrin receptor and mitochondria, so we proposed that besides neuronotrophic activity, bFGF might also be in charge with the modulation of metabolic function during the development.
