Enhancing the Polysaccharide Production of Cordyceps Sinensis by Using Aeration Strategy in Fermentor

In this research the effect of oxygen concentration and rate of aeration on the production of polysaccharides by C. sinensis was studied. A higher rate of sugar consumption and the inhibition of both cell growth and polysaccharide production were found under the high oxygen concentration cultivation of C. sinensis. The lowest extra-cellular polysaccharide (EPS) production was also found with 1 vvm aeration at 40% O2. On the other hand, after the cell grew 48 hr at 1 vvm aeration, then the aeration decreased to 0.5 vvm. A higher cell concentration and significant increase in polysaccharide production (2.8 g/L) were achieved. A lower molecular weight of extra-cellular polysaccharides was found in aeration with higher concentration of oxygen and a higher molecular weight of polysaccharides was found in lower aeration rate. For intercellular polysaccharide (IPS), a lower concentration of polysaccharidesl was also found in 1 vvm at 40% O2 and the highest IPS production was to be achieved a by using the aeration strategy of decreasing aeration at the middle stage of fermentation. The factors of dissolved oxygen concentration and the available carbon source played an important role in higher polysaccharides production. The results of this study can be used to enhance production of polysaccharides of C. sinensis and reduce the production cost.
