黑眼豇豆嵌紋病毒台灣分離株鞘蛋白基因與3' 端非轉譯區序列之解讀與分析

摘要


黑眼豇豆嵌紋病毒(Blackeye cowpea mosaic virus,BlCMV)屬於長絲狀的馬鈴薯Y病毒屬(Potyvirus)病毒,為豆科作物最重要病毒之一。利用反轉錄聚合?連鎖反應選殖黑眼豇豆嵌紋病毒之3'端區域,經譯讀後顯示其鞘蛋白基因共有861個核?酸,可轉譯成287個胺基酸,3'端非轉疫區部分則具有251個核?酸。由於鞘蛋白基因及3'端非轉譯區序列可作為Potyvirus屬鑑別與分類之標準,故本研究之主要目的為,藉分子層次上之比對,以明瞭BlCMV與幾種已發表的bean common mosaic subgroup病毒間之親緣性。比對結果顯示,台灣本地之BlCMV-TW分離株與國外報告之BlCMV-F(佛羅里達)、B1CMV-W、CAbMV-TH(泰國)、AzMV-H(日本)、PStV-ChinaG(中國)、PStV-B(美國)、BCMV-NY15(美國)、BCMV-NL1及BCMV-NL4彼此親緣關係極為接近,彼此之間相同度皆大於90%。因此這些不同地區報告之病毒應屬於同一病毒(B1CMV)的不同品系。其中台灣本地之B1CMV-TW鞘蛋白基因序列與B1CMV-F及CAbMV-TH具較高之相同度,與PStV-B及PStV-ChinaG具較低之相同度。而B1CMV之3’端非轉譯區則與B1CMV-W具有較高之相同度,與PStV-B具較低之相同度。

Blackeye cowpea mosaic virus (B1CMV) is a member of the genus Potyvirus and is one of the most important virus infecting leguminaceous crops. In this study, the cDNAs of the 3'-terminal regions of B1CMV-TW was amplified by RT-PCR and cloned. The nucleotide and amino acid sequences of coat protein (CP) gene and 3' non-translated region (NTR) of B1CMV-TW was analyzed and compared with other reported members of bean common mosaic virus subgroup (B1CMV-F、B1CMV-W、CabMV-TH、AzMV-H、PStV-B、BCMV-NY15、BCMV-NL1 and BCMV-NL4)from different geographic areas. The results showed that the sequence of 1112 bp located in the 3’terminal regions of BICMV-TW including 861 nucleotides of CP gene encoding a 287 amino acid which was followed by a non-translated region of 251 nuc1eotides and a 3' poly(A) tail. Comparison of BlCMV-TW with other reported members of bean common mosaic virus subgroup indicated that they are all the same virus but different strains. The results also showed that CP gene of BlCMV-TW had a higher identity with BICMV-F and CAbMV- TH, and a lower identity with PStV-B and PStV-China G. The 3' non-translated region of BICMV-TW has a higher identity with BlCMV-W, and a lower identity with PStV-B.
