台灣和金門地區螺旋線蟲(Nematoda: Rotylenchinae)之種類鑑定

Identification of Spiral Nematode (Nematode: Rotylenchinae) Collected from Taiwan and Kinmen

摘要


自2005年起,從台灣和金門地區不同地方和寄主作物的根圈土壤中分離得到9群helicotylenchus dihystera (Cobb,1893) Sher,1961、6群H.crenacauda Sher,1966,以及4群Rotylenchus brevicaudatus Colbran,1962。各群線蟲之種類鑑定依據包括外觀形態、形態測量值(morphometrics)及核糖體DNA (ribosomal DNA,rDNA)片段之5.8S基因和內轉錄區間(Internal transcribed spacer,ITS)的序列相同度比對分析。本研究提供更多完整的形態測量值和SEM外觀形態方面資料,並首次在GenBank基因庫中登錄這三種螺旋線蟲DNA片段(由4個序列以上所獲得之,包含5.8S基因、ITS-1及ITS-2),大小分別為1317bp、1316bp及1222bp之代表性序列,提供世界各地研究者一項重要的輔助鑑定資料。

Soil samples collected from the rhizosphere of various crops in several locations in Taiwan and Kinmen yielded nine populations of Helicotylenchus dihystera (Cobb, 1893) Sher, 1961, six populations of H. crenacauda Sher, 1966 and four populations of Rotylenchus brevicaudalus Colbran, 1962. Based on the external morphology, morphometrics and the sequence of internal transcribed spacer (ITS) region and 5.8S gene of the ribosomal DNA, the species taxon for each nematode population was confirmed. In this study, additional morphometric data and SEM figures for these three spiral nematode species were also provided. The sequence of representative rDNA fragment of each species was the first deposited in GenBank for the benefits of worldwide researchers.
