Molecular characterization of Broad bean wilt virus-2 isolated from lisianthus in Taiwan

自臺灣洋桔梗分離的蠶豆萎凋病毒-2之分子特性分析

摘要


中臺灣地區栽種的洋桔梗葉片顯現黃綠色環斑及壞疽病斑病徵,經過分離後得到一病毒分離株,命名為LV-5。在先前的報告中指出由於LV-5 可與市售的BBWV-2 抗血清反應,因此認為LV-5 可能屬於蠶豆萎凋病毒-2(Broad bean wilt virus-2, BBWV-2)的其中一個分離株。本研究的目標在於以分子生物學方式提供資料,以確定LV-5 病毒分離株為BBWV-2 的成員之一,並且將此病毒的全基因組加以選殖定序。從全基因組解序的資料得知:LV-5 分離株中除了病毒序列3 端的poly-A tail 以外,RNA 1 全長為5947 個核苷酸,RNA 2 為3555 個核苷酸。序列比對結果顯示LV-5 與其他7 個BBWV-2 分離株的大鞘蛋白(large coat protein, LCP)及小鞘蛋白(small coat protein, SCP)在胺基酸的序列相同度分別為84.8-97.5%及 90.9-98.0%。藉由RNA 1 及RNA 2 的全長核苷酸序列和兩個鞘蛋白的氨基酸序列的親緣關係分析結果顯示,從臺灣洋桔梗上所分離得到的LV-5分離株與日本甜椒上所分離的IP 分離株最為接近。綜合以上結果,本次研究提供從臺灣的洋桔梗上所得到的LV-5 分離株於分子生物學上的鑑定資料,確定LV-5 分離株為BBWV-2 的其中一個分離株。

A virus isolate LV-5 obtained from lisianthus (Eustoma russellianum) showing chlorotic and necrotic spots on leaves in central Taiwan, was previously identified as a potential isolate of Broad bean wilt virus-2 (BBWV-2) since it reacted positively with the commercially available BBWV-2 antiserum. To confirm this, the complete genomic sequences of LV-5 were determined and analyzed. LV-5 was identified to consist of 5947 nt for RNA 1 and 3555 nt for RNA 2, via genome walking. Sequence comparisons of the large coat protein (LCP) and small coat protein (SCP) showed that LV-5 shared 84.8-97.5% and 90.9-98.0% of amino acid identities, respectively, with those of seven other BBWV-2 isolates. Phylogenetic analyses based on the RNA polymerase and coat proteins revealed that lisianthus-infecting LV-5 was closely related to BBWV-2 IP, a Japan isolate. Our results demonstrated that virus LV-5 isolated from lisianthus in Taiwan is an isolate of BBWV-2. This is the first report of genome information of lisianthus-infecting BBWV-2.
