菸草嵌紋病毒矮牽牛分離株之特性分析

Characterization of the petunia isolate of Tobacco mosaic virus.

矮牽牛(petunia,Petunia x hybrida)植株具有疑似黃化和嵌紋病徵。經三次單斑分離後得一病毒株,以間接酵素結合抗體檢定法檢測,與菸草嵌紋病毒(Tobacco mosaic virus,TMV)抗體呈陽性反應。病毒株以TMV pet-TW稱之。TMV pet-TW 經機械接種可以感染茄科植物,並可在奎藜、紅藜、菸草、蕃茄、茄子、辣椒、千日紅及雞冠花等植物引起局部性病斑。病毒顆粒為直桿狀,大小約300 x 18 nm。病毒的基因體RNA 為單一分子,約6.4 kb。SDSPAGE顯示病毒鞘蛋白的相對分子量約為17.5 kDa,該蛋白於西方轉漬免疫分析時與TMV 之抗體有正反應。譯讀TMV pet-TW 的基因序列得全長序列含核苷酸6349 核苷酸(accession number: EF392659),其鹼基組成包含1883 個A(29.65%)、1218 個C(19.18%)、1495 個G(23.54 %)、1753 個T(27.61%)。TMV pet-TW 基因組RNA 有四個開放轉譯架構,分別為RdRp(RNA-dependent RNA polymerase)/replicase protein、RdRp/readthrough protein、MP(movement protein)和CP(coat protein)。比較TMV pet-TW 與他種菸草嵌紋病毒(TMV)各基因之核苷酸序列相同度及胺基酸序列相似度,結果顯示其確為菸草嵌紋病毒的矮牽牛分離株。核苷酸或胺基酸序列比對結果可知,TMV pet-TW(accession number: EF392659)與TMV 的B(AJ011933)、variant 1(V01408)、NC 82(X68110)、Fujian(AF395127)和Rakkyo(D63809)等分離株較為相近。研究結果亦顯示TMV pet-TW 之MP基因核苷酸和胺基酸序列數目與已知的異源TMV 分離株有差異。

Etiology of petunia (Petunia x hybrida) plants showing chlorosis and mosaic symptoms found in the nursery were investigated. A virus was isolated and found to react positively with antibody of Tobacco mosaic virus (TMV) in ELISA test, and thus was designated as TMV pet-TW. TMV pet-TW systemically infected some plants belonging to solanaceae, including petunia, eggplants, tomatoes, and Nicotiana benthamiana, whereas the virus caused local lesions on Chenopodium quinoa, C. amaranticolor, Nicotiana rustica, chili pepper, Gomphrena globosa and Celocea agentea. The particle of TMV pet-TW is rigid-rod in shape and abot 300 x 18 nm in size, consisting of a single molecule. SDS-PAGE analysis identified a single subunit of coat protein with a relative molecular weight of about 17.5 kDa. The CP subunit reacted positively with antibody of TMV indicating TMV pet-TW virus is a TMV-related isolate. Full-length genome of TMV pet-TW consists of 6349 nuclotides (accession number: EF392659) and encodes four open reading frames (ORFs), including a RNAdependent RNA polymerase (RdRp)/replicase gene, a RdRp/readthrough protein gene, a movement protein gene (MP) and a coat protein gene (CP). Comparison of nucleotide and amino acid sequences of TMV pet-TW to published TMV sequences revealed that TMV pet-TW displayed most similarity with TMV isolates of B (AJ011933), variant 1 (V01408), NC 82 (X68110), Fujian (AF395127) and Rakkyo (D63809).
