以酵母融合菌株Saccharomyces cerevisiae GSHOL2生產生質酒精與麩胱甘肽

A Study on Production of Bioethanol and Glutathione by Saccharomyces Cerevisiae GSHOL2

摘要


生質酒精(Bioethanol)係指以糖質、澱粉質或纖維質等生質為料源,利用微生物生物轉化而得,可摻配適當比例於石化汽油中,為汽車動力來源。麩胱甘肽(Glutathione, GSH)在臨床上常用於中毒性肝炎的治療、角膜與視網膜疾病的改善、有機物及重金屬的解毒、以及癌症放療及化療的保護等,亦廣泛用於化妝品工業及食品加工業等領域。本研究篩選出具生產生質酒精與GSH之酵母菌株Saccharomyces cerevisiae GSHOL2,並進行發酵生產生質酒精與GSH製程最適化探討。依序以饋料-批次式發酵生產生質酒精及GSH,分別於發酵生產生質酒精48小時後與繼續發酵生產GSH 48小時後,可得11.6% (wt%)生質酒精及18.7 mg/g-DCW GSH。添加適量過氧化氫(450 μM)可提高GSH在細胞內的濃度,但對細胞有傷害不宜在細胞生長穩定期前添加。本研究結果可增益附加經濟價值、降低總生產成本及作為商業化製程之參考。

Bioethanol is produced by the fermentation of sugar-based, starch-based, or cellulosic renewable materials, and is currently blended into gasoline as a source of green fuel. Glutathione (GSH) is usually used in treatment of toxic hepatitis, ameliorate diseases of cornea and retina, detoxification of organics and heavy metals, and protection on radiotherapy and chemotherapy of tumors. Nowadays, GSH is also widely used in cosmetic and food processing industry. This study has been screened a hybrid yeast strain, Saccharomyces cerevisiae GSHOL2, which can produce both of ethanol and GSH. Process optimization of fermentative productions of ethanol and GSH by Saccharomyces cerevisiae GSHOL2 has been discussed. The ca. 11.6% (wt%) ethanol and 18.7 mg/g-DCW GSH can obtain after producing GSH for 48 hrs followed by ethanol produced for 48 hrs with fed-batch and batch fermentation in sequence. On the contrary, intracellular concentration of GSH can be improved by suitable addition of hydrogen peroxide after stationary phase due to the reason that hydrogen peroxide is still harmful to yeast cells. The results can not only increase additional economic values but also reduce the total cost of production. Moreover, it can also be used as a reference for commercialization in the future.
