化學接枝改質聚丙烯不織布與膠原蛋白及幾丁聚醣固定對物化性及天竺鼠傷口癒合之研究

摘要


本研究係將聚丙烯不織布利用化學起始反應,將丙烯酸單體接枝於聚丙烯不織布上,實驗中利用戊二醛為交聯劑與不同比例之膠原蛋白與幾丁聚醣進行交聯反應,並固定於表面已改質之聚丙烯不織布表面,進而探討聚丙烯不織布經不同比例之膠原蛋白與幾丁聚醣固定後其物化性(固定率、FTIR官能基定性分析、吸水率)及抗菌性(菌落數、阻止帶)之變化,最後再應用於天竺鼠傷口癒合實驗,於不同時間下觀察傷口的癒合情形。由實驗結果得知,聚丙烯不織布經不同條件固定後,以戊二醛溶液用量為25g,固定時間18小時以內可得較佳的固定率,且固定率隨接枝率的增加而提升。抗菌性測試方面,聚丙烯不織布的抗菌效果(菌落數、阻止帶)隨著幾丁聚醣溶液比例的增加而提升,且以膠原蛋白溶液:幾丁聚醣溶液=2:3時具有100%的滅菌效果。最後由天竺鼠傷口的癒合程度可知,經膠原蛋白及幾丁聚醣固定後之接枝聚丙烯不織布具有優良的傷口癒合效果,可有效促進傷口的痊癒。

In this study, the grafting of acrylic acid onto polypropylene nonwoven fabric by the aid of benzoyl peroxide as a chemical initiator have been investigated firstly, then the cross-linking bonding of collagen and chitosan mixed with the surface of PP nonwoven fabric are performed by the cross-linker of glutaraldehyde. The effect of different graft yield to the immobilization ratio of collagen and chitosan mixed will be discussed, and the antibacterial capacity of different concentration of the chitosan, physical&chemical properties of PP nonwoven fabric immobilized with collagen and chitosan, skin wound of guinea pig all were discussed. The results indicated that the better immobilization ratio was attained with the glutaraldehyde of 25g, the reaction time of 18hr, and an increase in graft yield were found to increase the immobilization ratio. On antibacterial capacity, the 100% antibacterial capacity were attained in the ratio of 2:3(collagen: chitosan), and the antibacterial capacity will increase with increasing in the ratio of chitosan. Coverage of the wound area with the PP nonwoven immobilized with collagen and chitosan were significant smaller than PP nonwoven fabric after day 7, and completely healed by 21days. Therefore, PP nonwoven immobilized with collagen and chitosan is a safe biomaterial for use as a wound dressing for skin healing.
