鏻鹽化幾丁聚醣衍生物抗菌活性與細胞相容性之研究

Antibacterial Activity and Cytocompatibility of Phosphonium Chitosan Derivatives

摘要


本研究係以幾丁聚醣與N-環氧基2,3 丙基-N-甲基-N,N-二烯丙基甲基磺酸化銨 (MDAA)反應得到O-(2-羥基) 丙基-3-甲基-二丙烯基氨幾丁聚醣磺酸化物(O-HDACS),之後O-HDACS 分別與2 種N-(羧烷基)三苯基溴化鏻化合物(CATPP)進行醯氨化反應而得2 種鏻化幾丁聚醣衍生物(NQPCS)。NQPCS 的特性以傅立葉轉換紅外光光譜儀(FT-IR)分析其分子結構;以DPPH 測定其自由基抑制率。並以金黃色葡萄球菌(Staphylococcus aureus)測試最小殺菌濃度(MBC)。另外,細胞相容性以L929 纖維母細胞來評估其細胞成長活性與細胞毒性。

In this project, the O-(2-hydroxy) propyl-3-methyl diallyl ammonium chitosan salt (O-HDACS) were applied to make reaction of chitosan and N-epoxy-2,3-propyl-N-methyl-N,N-diallylammonium methyl sulfate (MDAA). Afterwards, two N-quaternary phosphoniun chitosan derivatives (NQPCS) were synthesized by O-HDACS and two (N-carboxyalkyl) triphenylphosphonium of compounds (CATPPs). These NQPCS were characterized by FT-IR. The antibacterial activities of NQPCS were tested by Staphylococus aureus. The antibacterial activity was evaluated by minimum bactericidal concentration (MBC). NQPCS antioxidant activity were evaluated by DPPH (2,2-diphenyl-1-picryhydrazyl) radical scavenging assay. In addition, the cytocompatibility was evaluated by L929 fibroblast proliferation and cytotoxicity.
