魚類分泌螢光物質至水中的檢測與特性分析

Detection and Characterization of Fish Secretory Fluorescence Substances

摘要


本研究旨在探討魚類表皮會分泌螢光物質至水中影響水質與研究此螢光訊號在不同物理環境下的特性。利用螢光光譜儀偵測蓄養斑馬魚的去離子水質溶解有機物時,意外發現魚類會分泌出具極強螢光訊號的物質(激發光350nm;放射光420nm),且斑馬魚胚胎、台灣鯛(吳郭魚)、鰻魚與青鱂魚亦具有分泌此螢光物質至水中之特性。以斑馬魚為模式生物研究顯示,魚類釋放螢光物質的訊號會隨著飼養時間延長而增加。利用蛋白酵素、加熱與改變酸鹼值後測定檢測結果顯示,魚類分泌至水中的螢光訊號具有極高的穩定度。魚體解剖試驗結果顯示此具螢光物質的物質主要是由魚表皮所分泌。利用HPLC分析,結果顯示魚類至少分泌4種具螢光訊號之物質至水中。此研究結果顯示魚類會分泌微量螢光物質至水中,同時提供利用高感度螢光光譜儀檢做為養魚水水質即時監測的新方法。

The experiments were studied on the detection and characterization of the aquatic water fluorescent substance which was secreted by fish. Fish health is extremely associated with the water quality of the environment, so fish skin mucous cells secrete mucus antimicrobial peptides (e.g. pleurocidin) against environmental stimuli and bacterial invasion. We found fish secrete the water-soluble fluorescent materials (excitation at 350 nm and emission at 420 nm) by high sensitive fluorescent spectrometry. The results of tissues dissection revealed that the fluorescent materials were mainly secreted from the fish skin. The fish water was treated with heat, salt, acid, base and proteinase, and the results revealed that the fluorescent signal was not altered after those treatments; it indicated that the fluorescent substances have a high stability. The results of HPLC analysis result revealed that the fish water contains at least 4 different fluorescent substances. We discovered that fish secretes these water-soluble fluorescent materials to water. This may be used as a novel protocol for the detection on the aquatic water quality.
