評估噬菌體對Xanthomonas campestris pv. campestris菌株之寄主範圍及裂解能力

摘要


由Xanthomonas campestris pv. campestris(Xcc)所引起的黑腐病,是十字花科作物的世界性重要病害。自台灣不同的十字花科蔬菜產區收集58株菌株,經剪葉接種及再分離等測試鑑定為致病株。之後利用專一性引子對進行多引子聚合酶連鎖反應,鑑定58株菌株皆為Xcc。而17株供試噬菌體則分離自十字花科蔬菜產區之罹病植株、田土及育苗場泥炭土,根據17株噬菌體對58株Xcc菌株的寄主範圍測試,可將17株噬菌體分為13群,其寄主範圍介於33-40Xcc菌株之間。寄主範圍最廣的第1群噬菌體可裂解40株供試Xcc菌株,最少的第12及13群噬菌體亦可裂解33株供試Xcc菌株,其寄主專一性與採樣年代、蔬菜產地及蔬菜種類無關聯性。選擇寄主範圍最廣的第1群噬菌體φXcc25進行溶菌能力測試,其在半固態或液態培養基中,分別降低83.8%或99.1%的寄主Xcc50菌量,顯示噬菌體φXcc25不論是在半固態或液態環境下,均能裂解Xcc菌株,且在液態環境表現較佳的溶菌能力。本研究顯示溶裂型噬菌體對甘藍黑腐病具有防治潛力。

Black rot caused by Xanthomonas campestris pv. campestris (Xcc) is a global important disease of cruciferous crops. Fifty-eight isolates were collected from different production areas of cruciferous vegetables in Taiwan, and identified as the pathogenic strains by leaf clipping inoculation and re-isolation tests. All 58 strains were identifi ed as Xcc by a multiplex polymerase chain reaction using the specifi c primer pairs. The 17 tested phages were isolated from diseased plants and fi eld soils in cruciferous vegetable production areas and nursery peat soils. Based on the host range of the 17 phages infective for 58 Xcc strains, the 17 phages were divided into 13 groups with a host range between 33 to 40 strains. The broadest host range, Group 1 phage, can lyse 40 Xcc strains, and the smallest Group 12 and 13 phages can lyse 33 Xcc strains. The host specifi city of phage was not associated with the sampling year, vegetable production area and vegetable species. The phage φXcc25, a Group 1 phage with the widest host range, was selected for the lysis ability test, and it reduced 83.8% or 99.1% of the host Xcc50 bacterial cells in semi-solid or liquid medium, respectively. Results indicated the phage φXcc25 can lyse Xcc strains in both semi-solid and liquid environments, and it implied better lysis ability in liquid environment. This study showed the potential of lytic phages for the control black rot of cabbage.
