非洲菊利用花蕾培養處理疊氮化鈉進行誘變育種

摘要


將非洲菊'Rosabella'、'Arobella'、以及'Rosalin'花蕾培養在含BA 5 mg/L及IAA 0.1 mg/L之1/2 MS基本培養基,其再生不定芽之能力,以緊蕾期(直徑 0.5~1.5 cm)最好;'Stardust'則以著色期的花蕾再生率較高。花蕾培殖體再生芽體所需時間,以'Rosabella'、'Arobella'以及'Foske'最短,需時3週,而'Ansofie'最長,需時7週。以1/2 MS基本培養基添加BA 5~10 mg/L培養'Twiggy'等五品種之花蕾,可再生較多芽體。'Rosabella'緊蕾期的花蕾以NaN3 0.5或1.0 mM浸漬60或90 min再生較多芽體。再生之變異株有花色變淡、花朵直徑變小、花瓣數變多等性狀的變化。

The best stage for shoot proliferation from flower buds of gerbera such as 'Rosabella', 'Arobella' and 'Rosalin', were at tight bud (0.5~1.5 cm in diameter), as well as 'Stardust' was at coloring stage. The shortest time for shoot regeneration required 3 wks in 'Rosabella', 'Arobella' and 'Foske', as compared to required 7 wks in 'Ansofie'. The 1/2MS medium containing BA 5~10 mg/L culturing 5 gerbera, such as 'Twiggy' proliferated more shoots. Tight buds of 'Rosabella' soaked in NaN3 0.5~1.0 mM for 60 or 90 min regenerated more shoots. Mutants of color, flower shape and number of petals were observed in the regenerated population.
