胚拯救培育抗黑腐病花椰菜與白菜種間雜種

Interspecific Hybrid between Brassica oleracea var. botrytis and B. rapa Developed through Embryo Rescue for Resistance to Black Rot

摘要


甘藍類(Brassica oleracea)黑腐病是由Xanthomonas campestris pv. campestris (Xcc)所引起,目前區分為9個生理小種,其中以生理小種1與4最主要。由於甘藍類缺乏兼抗黑腐病菌生理小種1與4的種原,限制了抗黑腐病育種的進展。此研究引進兼抗生理小種1與4的白菜類(Brassica rapa)種原PI 193847,與不同品系花椰菜(B. oleracea var. botrytis)雜交,摘取未成熟胚培育種間雜交種。胚培養成功率受母本基因型的影響,其正交與反交生育情形具有差異性。種間雜交苗經流式細胞儀檢定核酸含量介於兩親本間,確定為雜交後代,經利用噴霧接種法分別接種黑腐病原菌生理小種1與4,結果種間雜交種的罹病度為0-25%,呈高抗性。

Black rot of "Brassica oleracea" is caused by "Xanthomonas campestris pv. campestris" (Xcc). Classification of strains has been partitioned into 9 races and two race-typing (1 and 4) were predominant in "B. oleracea". The lack of resistant resources within germplasms has restrained resistance breeding of black rot in B. oleracea. In this study, resistant accession of "B. rapa" ( PI 173847) was introduced to cross with several lines of "B. oleracea" var. botrytis and the embryo culture techniques were used to culture immature embryo of interspecific hybrids. The efficiency of embryo culture was affected by the maternal genotype and the different stages of embryonic growth were caused by the reciprocal crosses. Flow cytometric analysis showed that interspecific hybrids had nucleic acid content between two parent lines. The results indicated that hybrid lines were highly resistant to Xcc races 1 and 4 after inoculation and the disease severity were between 0-25%.
