本研究調查台灣南部二仁溪水樣中酵母菌之種類與組成,共分離酵母菌八十二株,利用核醣體DNA(rDNA)中內間隔轉錄區(internal transcribed spacer, ITS)的限制酵素片段長度多型性(restriction fragment length polymorphism, RFLP)、rDNA中的LSU D1/D2片段與ITS區域的序列比對、Biolog菌種鑑定系統與形態特性鑑定菌種。河水酵母菌共區分出十五個RFLP菌群,鑑定出四屬八種,其中Pichia guilliermondii與Pichia ohmeri為台灣新紀錄種。在密度與分佈上,以Candida 屬佔優勢,紅色酵母菌稀少,應與二仁溪嚴重污染之水質特性有關。
This study investigated the yeast flora in the Er-Jen river in southern Taiwan. The Er-Jen river is mainly polluted by wastewater from industry and intensive animal feedlots. Yeasts were isolated, and their density was estimated. Fifteen yeast groups were clustered and eight species of four genera identified from 82 strains using ribosomal DNA (rDNA) internal transcribed spacer (ITS) region RFLP analysis, sequences of the rDNA LSU D1/D2 region and ITS region, morphological characters, and physiological tests performed using the Biolog yeast identification system. Pichia guilliermondii and Pichia ohmeri are reported for the first time in Taiwan. Major yeast populations were Candida. Candida spp. existed wildly, but little red/pink yeast presented, which correlated with human activity and river pollution.
