利用rRNA基因ITS 1、5.8S、ITS 2區域核酸序列鑑別冬蟲夏草(Cordyceps sinensis (Berk.) Sacc.)之研究

摘要


冬蟲夏草(Cordyceps sinensis (Berk.) Sacc.)自古為我國滋肺補腎的珍貴藥材。目前已確認冬蟲夏草無性世代為中國被毛孢(Hirsutella sinensis),但各不同產地冬蟲夏草的種源關係及種或品種的鑑定仍無法由目前的分類指標來區別。本試驗中以rRNA基因ITS 1(internal transcribed spacers 1)、5.8S、ITS 2區域序列進行冬蟲夏草類源關係的探討,由序列分析與基因庫資料比對的結果顯示冬蟲夏草可分成兩群,第Ⅰ群與H. sinensis (HMAS55469) ITS區序列相同度(identity)為98.9%;第Ⅱ群與H.sinensis (HMAS55469) ITS區序列相同度為61.8%。利用18SrRNA基因NS3/NS6區域核酸序列所確認同種的冬蟲夏草樣品,可進一步以ITS 1、5.8S、ITS 2區域核酸序列區分成兩群。

Cordyceps sinensis (Berk.) Sacc. is a treasured herb in Chinese medicine, a tonic for the lung and kidney. The anamorph of C. sinensis was identified as Hirsutella sinensis. But the identification of C. sinensis at the species level or below the species level using the traditional method based upon morphological observation is difficult. The phylogenetic relationship between C. sinensis samples was analyzed based on internal transcribed spacers 1 (ITS 1), 5.8S,and ITS 2 rRNA gene region sequences in this study. The results revealed that C. sinensis was divided into two clusters. The sequence identities of cluster Ⅰ and H. sinensis (HMAS55469) were around 98.9%. However, the sequence identity of cluster Ⅱ and H. sinensis (HMAS55469)was around 61.8%.The C. sinensis samples identified in the 18S rRNA gene NS3/NS6 regions at the species level could be further divided into two groups by using ITS 1, 5.8S, and ITS 2 rRNA gene sequences.
